A selective LZK (MAP3K13) inhibitor suppresses tumour growth and potentiates EGFR and PI3K pathway blockade in HNSCC and ESCC.

Strong contradiction

3 prior failures

Three or more documented clinical failures match this mechanism, or a Phase 3 efficacy failure is on record.

This preprint proposes a first-in-class LZK inhibitor and reports that it potentiates EGFR and PI3K pathway blockade in head and neck and esophageal squamous carcinoma. The Claidex graph holds three EGFR failures. tak-186-mvc-101-egfr-cd3-cobra-solid-tumors-phase1-2-efficacy-failure stopped a Phase 1/2 EGFR-directed T cell engager in a population that explicitly included head and neck squamous cell carcinoma, for limited anti-cancer activity. fpi-2107-egfr-cmet-nsclc-dosimetry-uptake-failure recorded a translational mismatch in which tumour uptake was limited by EGFR binding affinity. bg-60366-egfr-cdac-nsclc-strategic-shutdown closed a Phase 1 EGFR degrader on business grounds. Two of the three are efficacy or translational failures, which places this combination claim against the strongest prior-failure pattern the graph currently tracks for EGFR. The PI3K half of the proposal meets a second documented pattern, since alpelisib-epik-b3-tnbc and alpelisib-olaparib-pik3ca-ovarian-epik-o-phase3-efficacy-failure record two PIK3CA failures, one for efficacy and one for enrollment. Neither pattern bears on LZK itself, which has no failure history in the graph, so this flag applies to the proposed combination partners rather than to the novel target.

Abstract excerpt

Amplification of MAP3K13, which encodes leucine zipper-bearing kinase (LZK), occurs in a substantial proportion of head and neck squamous cell carcinoma (HNSCC) and esophageal squamous cell carcinoma (ESCC), highlighting LZK as an attractive therapeutic target. Among 75 compounds generated by systematic modification of the DLK inhibitor GNE-3511, compound #63 emerged as a highly potent LZK inhibitor with picomolar binding affinity. Pharmacologic inhibition of LZK by compound #63 suppressed kinase activity, reduced expression of the oncogenic effector c-MYC, and inhibited the viability of HNSCC and ESCC cells at low concentrations.
